Some people have found the GRAVE key (the button above the TAB key in North American keyboards) to be handy as a duplicate of the SPACEBAR, which closes all open windows so you can immediately address matters on the main screen (e.g., a pack of Elite Champions appeared and ambush your hero). Additionally, you can make your A or X keys duplicate the SHIFT key (attack while staying in one spot). You may find that binding your inventory (default I) to something like E or D might be handy. This way, you can spare your clicking for really deadly ambushes or attacks. One good way to save on hand fatigue is to bind a move key your character moves - but does not attack - to where your mouse points as long as the key is held.
